The presented kits are intended for isolation of DNA and RNA from plasma or serum samples.

DNA and RNA exist in plasma in different forms: in free form (very small percentage), included in exosomes and other extracellular vesicles (predominantly), in a complex with proteins and other macromolecules.

 

All the kits for DNA/RNA isolation with SileksMagNA or SileksMagNA-Direct particles are suitable for isolation of total nucleic acids from sample.

 

The kits can be used for isolation of viral RNA and DNA, mRNA, microRNA (isolation with SileksMagNA-Direct particles) and partly destructed or apoptotic nucleic acids.

When choosing the optimal kit for the isolation of nucleic acids from plasma and / or serum, it is necessary to determine what type of nucleic acid is planned to be isolated and how the isolated acid will be used after isolation.

 

1. The type of nucleic acid. DNA or RNA.

When RNA is isolated for subsequent analysis, a first strand synthesis reaction (reverse transcription) is usually required. In carrying out this particular reaction, impurities obtained during the isolation of RNA are of great importance.

The isolation on silicate particles of any manufacturer gives not the possibility to completely get rid of associated impurities. The silicate particles and the attached isolation system of any manufacturer will give some impurities that will affect the efficiency of the first strand synthesis reaction (reverse transcription), which will affect the results of further analysis.

The use of SileksMagNA-Direct particles for RNA isolation has a clear advantage in this case. This is due to the peculiarity of RNA binding on the particle surface and subsequent desorption. During these procedures, RNA is isolated as pure as possible, free of impurities. The sensitivity in the analysis of isolated RNA has the maximum value compared to other kits.

DNA is isolated as a rule for subsequent analysis in polymerase chain reaction (PCR). PCR and the enzymes involved in it are less sensitive to impurities. Therefore, the efficiency of DNA usage is less dependent on the purity of the isolation.

Consideration should be given to future use plans and the size of the nucleic acid to be secreted.

 

2. Further use.

SileksMagNA-Direct
The isolated RNA and DNA are recommended to be used immediately after isolation. Storage for more than 24 hours (even deep frozen) is not recommended.

The isolated RNA is recommended to be used immediately after isolation. Storage for more than 24 hours (even deep frozen) is not recommended.

The isolated DNA is recommended to be used immediately after isolation. Deep freeze storage possible.

 

3. The size of the nucleic acid to be isolated.

SileksMagNA-Direct
Isolated nucleic acids: single- and double-stranded nucleic acids, RNA and DNA, from 1-3 nucleotides to several thousand nucleotides. The efficiency of the isolation of very short and long nucleic acids is equal. Isolation of micro RNA is possible.

SileksMagNA
Isolated nucleic acids: single- and double-stranded nucleic acids, RNA and DNA, from 40-50 nucleotides to several thousand nucleotides. Double-stranded nucleic acids have an advantage in sorption.
